Will not boot up...

I am running Vista 64 home premium. In order for me to boot up the computer, I must have this blank formatted disk in the D drive. If it is not there the boot will take place and a black screen with most not all icons will appear. The start programs for the computer icon and control panel will work from the start programs box.
When the machine is turned on with the dvd disk inserted, it appears to be reading something from the disk as the active light lights up for about 5 or 6 seconds, just flickering away and then the desk top pops up, although everything seems to be much slower. All was working correctly at the beginning of the month and I do not know what I did wrong. At some time I did drag turbo tax and quicken onto the dvd but then I took both programs off. I would appreciate any help. Can't let the grandkids know I screwed up as they felt that I could not build the machine and that I was wasting their future money.
